General description

This Whole Effluent Toxicity (WET) proficiency testing sample is produced in accordance with ISO/IEC 17043:2010. This sample is manufactured according to TNI requirements. However, please note, this sample may include non-TNI analytes as well.

In acute WET tests, the small planktonic crustacean, Daphnia magna, an inhabitant of freshwaters, is employed as a test organism and exposed to various effluent concentrations under controlled conditions for a short duration (48 h). This helps to estimate the effluent′s toxicity and arrive at the lethally toxic concentrations of the effluent.

Preparation Note

Description

  • The effluent is provided as a concentrate for dilution in moderately hard synthetic freshwater (MHSF).
  • This concentrate is an EPA-specified toxicant in reagent-grade DI water.
  • Enough material has been provided to run the method at least twice. If you require more toxicant, please contact ptservice@milliporesigma.com.
  • Do not deviate from the supplied instructions or EPA Method 2021.0. Contact ptservice@milliporesigma.com before running the test if you see the need to deviate from the test conditions.
  • Deviations may affect your final evaluation.

Preparation Instructions


  • To prepare 1-L of effluent, add 900 mL of MHSF to a volumetric flask.
  • Preparation instructions for MHSF can be found in the EPA document EPA-821-R-02-012, page 32.
  • Transfer the entire contents of one ampule to the flask. Rinse the ampule with MHSF and transfer to the volumetric flask. Bring flask to 1 L volume with MHSF.
  • This represents 1 L of effluent water for test.


Required Test Conditions
  • Test Type: Static non-renewal
  • Test duration: 48 hours
  • Temperature: 25 ºC ± 1 ºC. The test must not deviate by more than 3 ºC during the test.
  • Renewal of Test solutions: Non-renewal
  • Dilution water: Moderately hard synthetic water


Test Conditions
  • Light quality: Ambient laboratory illumination (recommended)
  • Light intensity: 10-20 μE/m2 /s (50-100 ft-c) (ambient laboratory levels) (recommended)
  • Photoperiod: 16 h light, 8 h darkness (recommended)
  • Test chamber size: 30 mL (recommended minimum)
  • Test solution volume: 25 mL (recommended minimum)
  • Renewal of test solutions: After 48 h (required minimum)
  • Age of test organisms: Less than 24-h old (required)
  • No. of organisms per test chamber: 5 for effluent and receiving water tests (required minimum)
  • No. of replicate chambers per concentration: 4 for effluent and receiving water tests (required minimum)
  • No. of organisms per concentration: 20 for effluent and receiving water tests (required minimum)
  • Feeding regime: Feed YCT and Selenastrum while holding prior to the test; newly released young should have food available a minimum of 2 h prior to use in a test; add 0.1 mL each of YCT and Selenastrum 2 h prior to testing solution renewal at 48 h (recommended).
  • Test chamber cleaning: Cleaning not required
  • Test chamber aeration: None (recommended)
  • Test concentrations: Effluents: 5 and a control (required minimum)
